【摘要】:ThepresenceofcellgrowthmediumcomplicatesadirectanalysisofcellswithSIMS.Attemptstowashoutthenutrientmediumresultsintheexposureofcellstounphysiologicalreagentsthatmayalsoaltertheirchemicalcomposition.Thisobstacleisovercomebyusingasandwichfreeze-fracturemethod(1).ThiscryogenicmethodhasprovidedauniquewayofsamplingindividualcellsintheirnativestateforSIMSanalysis.TheproceduredescribedherehasbeensuccessfullyusedforimagingNa+andK+iontransport(3),calciumalterationsinstimulatedcells(4,5),andlocalizationoftherapeuticdrugsandisotopicallylabeledmoleculesinsinglecells(6).Thefrozenfreeze-driedcellspreparedaccordingtothismethodhavebeencheckedforSIMSmatrixeffects(7).Ionimagequantificationhasalsobeenachievedinthissampletype(8).Theproceduredescribedhereisamenabletoawidevarietyofcellculturesandprovidesawayforstudyingtheresponseofindividualcellsforchemicalalterationsinthestateofhealthanddiseaseandlocalizationofisotopically-labeledmoleculesandtheraputicdrugsincellculturemodels.1.1ThisguideprovidestheSecondaryIonMassSpectrometry(SIMS)analystwithacryogenicmethodforanalyzingindividualtissueculturecellsgrowinginvitro.Thisguideissuitableforfrozen-hydratedandfrozen-freeze-driedsampletypes.Includedareproceduresforcorrelatingoptical,laserscanningconfocalandsecondaryelectronmicroscopiestocomplementSIMSanalysis.1.2Thisguideisnotsuitableforcellculturesthatdonotattachtothesubstrate.1.3Thisguideisnotsuitableforanyplasticembeddedcellculturespecimens.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
